We chose to work on specified neurons with known properties and selected an invertebrate model preparation. Methods: Ganglionic neurons from Helix aspersa were exposed in a chamber containing physiological solution. Neurons were impaled with glass microelectrodes containing 0.7M KCl, K2SO4 or Kacetate. Current- and voltage-clamp recordings were made. Light was applied via an 80 m diameter optical fibre. Results: Neuron-specific effects on excitability were observed that resulted in each case from an increase in membrane conductance. Responses were intensitydependent, over the range of 25mW to 1200mW, reversible and repeatable. Some neurons were depolarized and excited, with an extrapolated Erev of + 60mV of the current response. Others were inhibited, with an Erev close to the resting Em. Detailed experiments on the C1 neuron, showed that inhibition resulted from an increase in Cl- ion conductance, that occurs most probably along the axon. Conclusions: 532 nm light consistently excited some neurons, probably by increasing membrane permeability to Na + and or Ca2 + ions, but inhibited the C1 neuron by a specific action on Cl- ion channels, for instance, solubility of griseofulvin.

The emphasis has been on block copoly oxyalkylene ; s which form spherical or near spherical micelles on dissolution in water. However, spherical micelles of copolymers with short E-block lengths relative to the length of the hydrophobic core-forming ; block may undergo a sphere-to-cylinder transition on heating the solution, an effect which has been recognized for certain EmPnEm copolymers for many years, e.g. for copolymers E27P39E27 denoted P85 ; and E21P47E21 denoted P94 ; see, e.g., [1924] ; . As indicated by Nagarajan [16], there is an advantage so far as solubilization capacity is concerned in using copolymers which form cylindrical micelles and we have synthesized copolymers which form cylindrical, even worm-like, micelles at room temperature or just above [2527]. In our work, all solutions were prepared by dissolving the copolymers directly in water at room temperature and, since values of the glass transition temperature Tg ; of the hydrophobic blocks are low, it can be safely assumed that all results are for systems at equilibrium. Of the components of the copolymers considered in this chapter, only poly styrene oxide ; and poly phenyl glycidyl ether ; have values of Tg above 0 8C, namely ca. 40 and 18 8C, respectively [2830], but these values apply to lengthy chains and Tg is much lower for the short blocks of the copolymers, as demonstrated directly for S blocks by investigation of chain dynamics in micellar cores [31]. Ss False-Positive Versus True-Positive Drug-Drug Interactions With Warfarin Warfarin is a marvelous, life-saving drug in the prevention of atherothrombotic events such as myocardial infarction, stroke, venous thromboembolism, and deep vein thrombosis. However, warfarin has somewhat unpredictable anticoagulant effects that require continuous monitoring of prothrombin time and the international normalization ratio INR ; , and the search continues for a drug with efficacy comparable to warfarin but with lower risk of over- and underanticoagulation.1 The need for continuous monitoring of warfarin is magnified by the potential for drug-drug interactions. Today, there are 225 drug-drug interactions listed in Facts and Comparisons for warfarin, of which 79 are categorized as a Level 1 interaction, the highest severity level.2 A Level 1 interaction is described as: "Potentially severe or life-threatening interaction; occurrence has been suspected, established or probable in well-controlled studies. Contraindicated drug combinations may also have this number." Among women sociodemographic characteristics are associated with differences in health and functional status. Because of sample size limitations, prior studies have been unable to adequately assess the independent effects of, and interactions between education, race ethnicity R E ; , and income as measures of socioeconomic status SES ; and health status in older women. We assess these factors using data from the 1999 Medicare Health Outcomes Study. The survey randomly sampled 1000 enrollees from each Medicare managed care plan in the US and had a response rate of 66%. Our analysis includes 87, 220 women age 65 and older including 5907 African American AA ; , 4376 Latina, and 1394 Asian women. To assess HRQOL we used the two SF-36 summary scores PCS and MCS ; and the 8 subscales of the SF-36. The contribution of comorbidity, measured by a count of self-reported chronic conditions, was assessed. In bivariate analysis there were significant differences in SES, comorbidity, and SF-36 scores by R E. 27% white, 30% Asian , 49% AA, and 55% Latina women reported HS education. Mean SF-36 scores were highest for Asian and lowest for AA women. In multivariable models age, income, education, and R E were independently associated with SF36 summary scores and subscales which was explained in part by greater comorbidity among women with lower SES. One aim of our research is to elucidate the nature and mechanism of formation of MBs, which consist of keratins, heat-shock proteins and ubiquitin. Moreover, we recently found a new protein, p62, associated with MBs, and will investigate its significance in the etiology of liver diseases and its role in cellular defence to ROS-mediated injury. Both mitochondrial production of ROS and damage caused by exposure to them is of importance: they lead to changes in mitochondrial and cytosolic redox-status as indicated by the ratios of oxidatively generated cardiolipin metabolites and the ratio of reduced and oxidized glutathione, respectively. Hence they will serve as important markers for the status and progression of such liver diseases. Introduction and previous work: A central topic of our research is the elucidation of the nature and pathogenesis of protein aggregates Mallory bodies, MBs ; arising in hepatocytes in the course of several chronic toxic and metabolic diseases e.g. alcoholic and non-alcoholic steatohepatitis, chronic cholestasis, chronic copper toxicity, diabetes mellitus and other disturbances of lipid metabolism ; in order to understand crucial pathogenetic mechanisms involved in cell damage associated with these disorders. MBs consist of keratins and non-keratin components, including heat shock proteins and ubiquitin. Moreover, recently a novel protein, p62, was found by our team to be associated with MBs. P62 is involved as adapter in signal transduction pathways e.g. TNF and IL-1-signaling leading to NFkB activation ; as well as in the sequestration of ubiquitinated proteins. Its significance in pathologic conditions, particularly in protein aggregation diseases, is still enigmatic and its analysis is a major goal of our work. Studies on the pathogenesis of MBs and associated cell damage are facilitated by animal models, in which these inclusion bodies can be reproduced under standardized experimental conditions griseofulvin- or DDCfeeding of mice ; . Experiments with keratin knock-out mice disclosed that inclusion body formation represents a novel protective response operative in the cell in addition to restoration of protein structure by the action of chaperones or proteolytic degradation of potentially harmful abnormal proteins. Application of modern techniques of genomics and proteomics e.g. knock-out mice, gene expression profiling, and tissue micro array technology ; will help to unravel the complex mechanisms and disturbed cellular networks involved in these disorders. Another important aspect in the pathogenesis of liver disease is oxidative stress resulting from mitochondrially generated reactive oxygen species ROS ; . The formation of superoxide and hydrogen peroxide lead to increased formation of oxidized lipids, such as cardiolipin CL ; , in the inner mitochondrial membrane IMM ; . CL is unique constituent of the mitochondrial inner membrane and contains a large percentage of the oxidizable lipids in IMM. It has been shown recently, that the CL-cytochrome c adduct in IMM serves as a specific peroxidase which is dependent on mitochondrial hydrogen peroxide. Moreover, since CL oxidation is prevented by antioxidants, and metabolites of CL can serve as specific indicators of the.
